      目的:探讨跑台运动训练对脊髓损伤(SCI)后大鼠肺功能及高迁移率族蛋白B1 (HMGB1)表达的影响。方法:选取96只SD雌鼠,随机分成4组,即正常组、假手术组、脊髓损伤非训练组、脊髓损伤训练组。脊髓损伤训练组大鼠于术后第3天开始进行跑台运动训练。分别在术后第3天、术后第14天检测4组大鼠动脉血气分析,肺组织湿/干重比值、大鼠肺损伤评分及肺组织中HMGB1的表达情况。结果:动脉血气分析结果显示术后第14天,脊髓损伤训练组血氧分压值(PaO2)显著高于SCI非训练组、二氧化碳分压值(PaCO2)显著低于SCI非训练组(P<0.05);术后第14天SCI训练组肺组织湿/干重比值较SCI非训练组明显降低(P<0.05)。肺组织HE染色结果显示,与SCI非训练组相比,术后第14天SCI训练组肺损伤评分明显降低(P<0.05)。免疫组化结果显示SCI非训练组与SCI训练组术后第3天肺组织中HMGB1表达水平均显著增加(P<0.05);术后第14天,与SCI非训练组相比,SCI训练组HMGB1表达水平显著降低(P<0.05)。结论:跑台运动训练可改善脊髓损伤后大鼠肺功能障碍,其机制可能与抑制HMGB1的表达有关。

      Objective: To explore the effect of treadmill training(TT) on lung function and high-mobility group box-1 protein (HMGB1) expression in rats after spinal cord injury(SCI). Methods: Ninety-six SD female rats were randomly divided into four groups: normal group, sham operation group, SCI group and SCI+TT exercise group. The rats in SCI+TT group began TT on the 3rd day after operation. Arterial blood gas analysis was dpme, and lung wet/dry weight ratio (wet/dry), lung injury score and the expression of HMGB1 in lung tissue were measured on the 3rd day and 14th day after operation. Results: Arterial blood gas analysis showed that PaO2 in SCI+TT group was significantly higher than that in SCI group and PaCO2 was significantly lower than that in SCI group on the 14th day after operation, and wet/dry in lung tissue in SCI+TT group was significantly lower than that in SCI group on the 14th day after operation. The HE staining results showed that compared with the SCI group, the lung injury score of the SCI+TT group was significantly reduced after 14 days (P<0.05). Immunohistochemical results showed that the level of HMGB1 expression in the lung tissues of the SCI group and the SCI+TT group increased significantly after 3 days (P<0.05). Compared with the SCI group, the expression level of HMGB1 of the SCI TT group significantly decreased at 14 days after operation (P<0.05). Conclusion: TT can improve lung dysfunction in rats with SCI in the thoracic segment, and the mechanism may be related to the inhibition of HMGB1 expression.
