  目的:观察针灸对SAMP8小鼠行为学及海马区β淀粉样蛋白1-42(Aβ1-42)、细胞内微管相关蛋白Tau(Tau)以及神经元特异性核蛋白(NeuN)三大主要病理特征的影响。方法:6月龄雄性SAMP8小鼠随机分为模型组(n=6)和针灸治疗组(n=6),以SAMR1雄性小鼠作为对照组(n=6)。对针灸治疗组针刺百会和双侧神门,艾灸双侧肾俞共28d。治疗后,采用Morris 水迷宫评价动物的学习记忆能力,免疫组化法检测Aβ1-42和NeuN蛋白表达水平,免疫蛋白印迹法检测Tau蛋白水平。结果:治疗结束24 h后,与模型组相比,从第2天开始,针灸治疗组逃避潜伏期明显缩短(P<0.01),原平台象限停留时间增加(P<0.05),穿越平台次数增加(P<0.05),海马Aβ1-42阳性表达减弱(P<0.01),海马NeuN阳性表达增强(P<0.05),海马Tau蛋白表达量降低(P<0.05)。结论:针灸百会、神门、肾俞穴能明显提高SAMP8小鼠学习记忆能力,可能与减少Aβ1-42沉积、降低Tau蛋白水平、抑制神经元数量减少有关。

  Objective:To explore the effects of acupuncture at GV20, HT7, and BL23 on the behavior, expression of β amyloid protein 1-42 (Aβ1-42), Tau protein and neuronal nuclei (NeuN) in hippocampus of SAMP8 mice. Methods:Six-month-old male SAMP8 mice were randomly divided into model group (n=6) and acupuncture treatment group (n=6), and SAMR1 male mice served as control group (n=6). Acupuncture treatment group was acupunctured at GV20 and HT7, then subjected to moxibustion on both sides of BL23 for 28 days. Morris water maze was used to evaluate the learning and memory ability of animals. The expression of Aβ1-42 and NeuN was detected by immunohistochemistry. Western blotting was used to detect the level of Tau protein in the hippocampus of mice. Results:As compared with the control group, escape latency was significantly prolonged (P<0.01), the residence time in the original platform quadrant was shortened (P<0.01), the number of crossing platforms was decreased (P<0.01), and the expression of Aβ1-42 was up-regulated (P<0.01), the expression of NeuN down-regulated (P<0.01), and the relative expression of hippocampal Tau protein up-regulating (P<0.01) in the model group. As compared with the model group at the second day, escape latency was shortened (P<0.01), the original platform quadrant stay time prolonged (P<0.05), the number of crossing platforms increased (P<0.05), the expression of hippocampal Aβ1-42 down-regulated (P<0.01), the expression of NeuN up-regulated (P<0.05) and the relative expression of hippocampal Tau protein down-regulated in the acupuncture treatment group. Conclusion:Acupuncture at GV20, HT7, and BL23 points can improve the learning and memory abilities of SAMP8 mice probably by reducing Aβ1-42 deposition and Tau protein levels, and inhibiting the decrease in the number of neurons.
