      目的:观察按经选穴针刺对失眠大鼠下丘脑5-HT 1a、5-HT 2a受体mRNA表达的影响,探讨针刺治疗失眠症的疗效机制及按经选穴针刺对腧穴配伍效应的影响。方法:将60只SD大鼠随机分为5组,即:空白组、模型组、百会+神门穴组、百会+三阴交穴组、百会+非经非穴组,每组12只,通过腹腔注射DL-4-氯苯基丙氨酸混悬液建立失眠模型大鼠,各治疗组针刺相应腧穴,每次30min,治疗7d。运用实时荧光定量方法检测大鼠下丘脑5-HT 1a、5-HT 2a 受体mRNA表达量。结果:与模型组比较,各针刺组大鼠下丘脑5-HT 1a受体mRNA表达量均有一定程度的升高,5-HT 2a受体mRNA表达量均有一定程度的降低,且以百会+神门穴组调节效果最明显,百会+三阴交穴组及百会+非经非穴组次之。结论:针刺治疗可能通过调节失眠大鼠下丘脑5-HT 1a、5-HT 2a受体mRNA的表达发挥治疗作用,且按经选穴针刺可能是影响腧穴配伍效应的影响因素之一。
英文摘要:
      Objective: To observe the effect of acupuncture treatment on 5-HT 1a and 5-HT 2a mRNA expression in hypothalamus of insomnia rats and to explore the effect of acupuncture on insomnia and investigate whether acupuncture acupoints at different meridians are the key factor to determine the difference of acupoints synergy effect. Methods: Sixty SD rats were randomly divided into 5 groups, including blank group, model group, Baihui + Shenmen group, Baihui + Sanyinjiao group, Baihui + non-acupoint group, with 12 rats in each group. Chlorophenylalanine suspension was injected intraperitoneally to establish insomnia model in rats. Each treatment group received acupuncture in relevant acupoints, 30 min each time, for 7 days. Real-time fluorescence quantitative PCR was used to detect the expression of 5-HT 1a and 5-HT 2a mRNA in hypothalamus of rats. Results: As compared with the model group, the expression of 5-HT 1a in the hypothalamus of each acupuncture group increased to a certain extent and the expression of 5-HT 2a decreased to a certain extent. The curative effect of Baihui + Shenmen group was the best, followed by Baihui + Sanyinjiao group. Conclusion: The therapeutic effect of acupuncture may be performed by regulating the expression of 5-HT 1a and 5-HT 2a mRNA in the hypothalamus of insomnia model rats. Acupuncture acupoints at different meridians may be one of the factors that cause the difference in acupoints synergy effect.
